In vitro study of prolonged-release formulations containing amoxicillin and Gelucire 64/02

Abstract

An option for obtaining prolonged-release formulations is fluid-bed coating of immediate action granulates with fatty excipients such as Gelucire. This method has been evaluated for an active ingredient of pH-dependent solubility such as amoxicillin. In particular, the influence of the pH of the dissolving medium and the proportions of active ingredient and excipient on the rate and mechanism of amoxicillin release has been studied. The use of the non-linear dissolution coefficients k and n is also analyzed.
